Reference Values for Peripheral Blood Lymphocyte Subsets of Healthy Adults by Two-color Direct Immunofluorescence Method


Abstract
Background :
@EN Lymphocyte subsets in peripheral blood are enumerated to assess the immune status and to diagnose various disease processes. Recently flow cytometry is used widely in routine clinical laboratories and now two-color direct immunofluorescence

cytometric analysis is the standard method for lymphocyte subset measurement. But only few datas about reference values in Koreans have been reported.
@ES Methods :
@EN Lymphocyte subset analysis was performed on 167 healthy adults using twocolor direct immunofluorescence flow cytometric analysis. Lymphocyte subsets analysed were T (CD3) and B (CD19) cells, helper/inducer (H/I) T (CD4) and
suppressor/cytotoxic
(S/C) T (CD8) cells, helper/suppressor (H/S) ratio, activated T cells (CD3HLA-DR), and natural killer (NK) cells (CD3CD16 and/or CD56). Seventy-eight samples were split and two-color stain for CD3CD4 and CD3CD8 were also used for the analysis of
H/I T
and S/C T cells.
@ES Results :
@EN The mean values of lymphocyte subsets were: total T cells 67.8%, B cells 13.9%, H/I T (CD4) cells 42.4%, S/C T (CD8) cells 33.9%, H/S ratio 1.33, activated T cells 14.4% and NK cells 18.6%. Female showed higher values of percentage of T
cells,
H/I T
cells and H/S ratio and the vales of percentage of H/I T cells, H/S ratio and activated T cells increased with age, Measurements of H/I T (CD3CD4) cells and S/C T (CD3CD8) cells using different antibody combinations showed significant differences
especially in the percentage of S/C T cells, and thus in H/S ratio.
@ES Conclusions :
@EN In the analysis of lymphocyte subset our data will serve as the reference values in adults, and the measurement of CD3CD4 cells and CD3CD8 cells is considered to be more useful for H/I T cells and S/C T cells than CD4 and CD8 cells(Korean J
